«Eerste stappen van glycolyse
Het eerste enzym dat in de tweede fase van glycolyse wordt gebruikt, is het glyceraldehyde 3-fosfaatdehydrogenase; dehydrogenasen zijn enzymen die de overdracht van reducerend vermogen katalyseren van een reducerend molecuul dat oxideert naar een ander gereduceerd molecuul (redoxreactie).De substraten van dit enzym zijn NAD (nicotidamide-adenine-dinucleotide) en FAD (flavine-adenine-dinucleotide).
In deze stap katalyseert de dehydrogenase de omzetting van glyceraldehyde 3-fosfaat in 1,3-bisfosfoglyceraat: op dezelfde katalytische plaats wordt de aldehydegroep geoxideerd tot carboxyl met als gevolg reductie van NAD + tot NADH en vervolgens kan de carboxylgroep om een anhydridebinding te vormen met een orthofosfaat. Het eerste proces is erg exergonisch (er komt energie vrij), terwijl het tweede erg exergonisch is (er is energie voor nodig); als er geen katalytische plaats zou zijn, zou de globale reactie niet plaatsvinden: de eerste reactie zou plaatsvinden met het vrijkomen van energie die zou worden verspreid als warmte en die daarom niet bruikbaar zou zijn om de anhydridebinding te vormen.
Na de vorming van 1,3-bisfosfoglyceraat hervat het enzym zijn uitgangsstructuur en is het klaar om op een nieuw substraat in te werken.
Dan komt de fosfoglyceraatkinase die de overdracht van een fosforyl van 1,3-bisfosfoglyceraat naar een ADP-molecuul mogelijk maakt; we hebben ATP verkregen (één ATP voor elk molecuul glyceraldehyde-3-fosfaat, dus twee ATP's voor elk aanvankelijk glucosemolecuul) dat het energieverbruik van de eerste fase van glycolyse.
Het arsenaatanion (AsO43-) beïnvloedt de glycolytische route omdat het het fosfaat kan vervangen in de eerste reactie van de tweede fase van de glycolyse, waardoor 1-arsenio-3-fosfoglyceraat ontstaat dat zeer onstabiel is en, zodra het vrij is van de katalytische site, hydrolyseert het vrijgeven van het "arsenaat dat terugkeert naar de bloedsomloop. Daarom bootst het arsenaat de werking van het fosfaat na en komt het in de katalytische plaats: in aanwezigheid van het arsenaat vindt de reactie die ATP produceert (van 1,3-bisfosfoglyceraat tot 3-fosfoglyceraat) niet plaats omdat het 3-fosfaatglyceraldehyde wordt direct omgezet in 3-fosfoglyceraat; als er geen ATP beschikbaar is, sterven cellen af (arseenzuurvergiftiging).
In de derde reactie van de oxidatieve fase wordt 3-fosfoglyceraat omgezet in 2-fosfoglyceraat door de werking van fosfoglyceraatmutase; de reactie omvat een 2,3-bisfosfoglyceraattussenproduct.
In de volgende stap grijpt een enzym in enolase dat in staat is de eliminatie van een watermolecuul uit het koolstofhoudende skelet van het 2-fosfoglyceraat te katalyseren, waardoor het gepyriveerde fosfoenol (PEP) wordt verkregen;
PEP heeft een hoog potentieel voor de overdracht van een fosforyl: het wordt overgedragen door de werking van een enzym pyruvaatkinase, een fosforyl tot een ADP om ATP te geven, in de vijfde stap van de tweede fase, waarbij pyruvaat wordt verkregen.
2-fosfoglyceraat en 3-fosfoglyceraat hebben een laag overdrachtsvermogen van een fosforyl, daarom wordt, om ATP uit deze moleculen te verkrijgen, 3-fosfoglyceraat tijdens glycolyse omgezet in 2-fosfoglyceraat, omdat het uit de laatste wordt verkregen. soorten met een hoog overdrachtspotentieel.
Laten we, voordat we verder gaan, een haakje openen over 2,3-bisfosfoglyceraat; de laatste is aanwezig in alle cellen waarin glycolyse plaatsvindt in een zeer lage concentratie (het is het tussenproduct van de derde reactie van de tweede fase van glycolyse). In erytrocyten daarentegen heeft 2,3-bisfosfoglyceraat een stationaire concentratie van 4-5 mM (maximale concentratie) omdat ze een enzymatisch patrimonium bezitten dat de taak heeft om het te produceren; in erytrocyten is er een afwijking van glycolyse om 2,3-bisfosfoglyceraat te produceren: 1,3-bisfosfoglyceraat wordt omgezet in 2,3-bisfosfoglyceraat door de werking van bisfosfoglyceraatmutase (erytrocyt) en 2,3-bisfosfoglyceraat, door de werking van bisfosfoglyceraatfosfatase (erytrocyt) wordt 3-fosfoglyceraat. Vervolgens wordt in de erytrocyten een deel van het 1,3-bisfosfoglyceraat verkregen uit glycolyse omgezet in 2,3-bisfosfoglyceraat dat vervolgens terugkeert naar de glycolytische route als 3-fosfoglyceraat; daarbij wordt de derde stap van de oxidatieve fase van de glycolyse waaruit ATP wordt verkregen De hoeveelheid ATP die verloren gaat is de prijs die een erytrocyt bereid is te betalen om de concentratie van 2,3-bisfosfoglyceraat te behouden die deze cellen nodig hebben omdat het het vermogen van "hemoglobine om zuurstof te binden" beïnvloedt.
We hebben gezien dat in de eerste reactie van de tweede fase van glycolyse de NAD+ wordt gereduceerd tot NADH maar het is noodzakelijk dat, na het verkrijgen van het pyruvaat, de NADH weer wordt omgezet in NAD+: dit gebeurt met melkzuurfermentatie (lactaat wordt verkregen) of door alcoholische fermentatie (pyruvaatdecarboxylase dat pyruvaat decarboxylaat en een dehydrogenase dat ethanol vormt spelen een rol); bij fermentaties is geen zuurstof betrokken (anaëroben).
Als gevolg van melkzuurfermentatie hoopt melkzuur, als het niet op de juiste manier wordt afgevoerd, zich op in de spieren en veroorzaakt het onwillekeurige spiersamentrekkingen en dus krampen door H+ vrij te geven; een spier die onder zware stress staat, kan ook een minimale pH van 6,8 bereiken.
Via de Cori-cyclus wordt een deel van de vermoeidheid van een spier overgebracht naar de lever wanneer de spier overbelast is. Stel dat de spier werkt zonder zuurstoftoevoer (verkeerde aanname): als de spier matig werkt, wordt de ATP die nodig is voor contractie uitsluitend geleverd door glycolyse.Als de activiteit van de spier toeneemt en extra ATP nodig is, versnelt u het aerobe metabolisme, lactaat, dat zo wordt afgevoerd, in glucose. In werkelijkheid maakt de spier gebruik van het aërobe metabolisme: als er zuurstof beschikbaar is, benut de spier vooral de ATP die wordt geleverd door het aërobe metabolisme en als er geen zuurstof meer beschikbaar is, wordt het anaërobe metabolisme versneld door de Cori-cyclus Deze cyclus gaat ervan uit dat lactaat wordt overgebracht van de spier naar de lever, waar, door energie te verbruiken, meer glucose wordt geproduceerd die terugkeert naar de spier. Door deze cyclus wordt een deel van de in de spier verbruikte ATP geleverd door de lever, die, door het gluconeogeneseproces, is in staat glucose te produceren die door de spier kan worden gebruikt om ATP te verkrijgen.
Het tot nu toe beschreven glucosemetabolisme omvat geen zuurstof, maar het aerobe metabolisme van glucose maakt het mogelijk om 17-18 keer hogere hoeveelheden ATP te verkrijgen dan die verkregen met de glycolytische route, dus wanneer de cel de mogelijkheid heeft om te kiezen tussen aerobe en ed anaëroob, begunstigt de eerste.
Bij aëroob metabolisme komt pyruvaat de mitochondriën binnen waar het transformaties ondergaat en uiteindelijk koolstofdioxide en water worden verkregen; op deze manier worden 34 moleculen ATP verkregen voor elk molecuul afgebroken glucose.
